RESUMO

We prepared a naturally occurring flavanoid namely quercetin from tea leaves and analyzed by Absorption, Emission, FT-IR, 1H, 13C nmr spectra and ESI-MS analysis. The inclusion behavior of quercetin in cyclodextrins like α-, ß-, γ-, per-6-ABCD and mono-6-ABCD cavities were supported such as UV-vis., Emission, FT-IR and ICD spectra and energy minimization studies. From the absorption and emission results, the type of complexes formed were found to depend on stoichiometry of Host:Guest. FT-IR data of CD complexes of quercetin supported inclusion complex formation of the substrate with α-, ß- and γ-CDs. The inclusion of host-guest complexation of quercetin with α-, ß-, γ-CDs, per-6-ABCD and mono-6-ABCDs provides very valuable information about the CD:quercetin complexes, the study also shows that ß-CD complexation improves water solubility, chemical stability and bioavailability of quercetin. Besides, phase solubility studies also supported the formation of 1:1 drug-CD soluble complexes. All these spectral results provide insight into the binding behavior of substrate into CD cavity in the order per-6-ABCD > Mono-6-ABCD > γ-CD > ß-CD > α-CD. The proposed model also finds strong support from the fact with excess CD this exciton coupling disappears indicates the formation of only 1:1 complex.

The establishment of drug product stability and sameness is the heart of generic formulation development. For regulatory filing, various instrumental methods are used on a case basis to establish the generic and innovator product sameness in multiple aspects. Here in the present study, we explored the applicability of the Time-correlated single photon counting (TCS-PC) technique as a fast, reliable, and nondestructive method for establishing the sameness of three different categories of injectable formulations, namely, Amphotericin B liposome for injection, enoxaparin injection, and iron sucrose injection. All three category formulations were evaluated in their native and artificially induced post degradation state to identify the discrimination power of the used instrumental techniques. The degradation of materials were confirmed by high performance liquid chromatography (HPLC). Based on the product category, pre and post-degradation samples were evaluated by selective instrumental methods like differential scanning calorimetry (DSC), nuclear magnetic resonance (NMR), fluorescence spectroscopy, particle size analysis by dynamic light scattering (DLS), small angle X-ray scattering (SAXS), Raman spectroscopy, inductively coupled plasma optical-emission spectrometry (ICP-OES) and circular dichroism study. All pre and post-degradation samples were further analyzed by TCS-PC. We observed that, TCS-PC can identify the differences between the initial and post degradation samples in very less time with promising discrimination power across the different injectable formulation types. Thus TCS-PC can be used as a fast and promising stability or sameness evaluation tool for different injectable drug products.

Synthesis of luminescent metal cluster for selective sensing of specific analyte with detail mechanistic understanding is very important for real world applications as well as for developing new emissive materials. In the present work, we have synthesized L-glutathione stabilized gold (Au-SG) and gold-silver bimetallic (AuAg-SG) clusters under identical experimental conditions with orange red emissive characteristics for both. Detail photo physical analysis reveals that both clusters are phosphorescent in nature with moderate quantum yield of 7% and 19% for Au-SG and AuAg-SG respectively and their excited state lifetime values are in the range of 1-2 µs. While Au-SG cluster showed luminescence quenching response (turn-off) in presence of Fe3+ and Hg2+ ions, AuAg-SG cluster showed turn-off response for Cu2+, Fe3+ and Hg2+, but luminescent enhancement (turn-on) response for Cd2+ ions. The highest detection limit obtained for Cu2+ ion by AuAg-SG cluster is 20 nM while for Cd2+ ion it is 75 nM. From Time Correlated Single Photo Counting (TCSPC) and Dynamic Light Scattering (DLS) measurements we postulated that except Cd2+, all other metal ions cause aggregation of clusters through ligation with SG ligands while Cd2+ ion does not induce any cluster aggregation but binds to cluster surface atoms. The near constant life time values of both clusters during gradual addition of respective metal ions confirms static quenching/enhancement process through formation of stable ground state adducts.

A pure water soluble xylan (PS-I) with an average molecular weight ~1.1 × 105 Da was isolated from the hot water extraction of fresh leaves of Piper betle (paan). The xylan was found to be composed of xylose, galactose and methyl galacturonate in a molar ratio of nearly 3:1:1. The repeating unit was composed of a backbone containing three (1 â†’ 4)-α-D-Xylp residues, one of which was branched at O-2 position with the side chain consisting of (1 â†’ 4)-α-D-GalpA6Me and terminal ß-D-Galp residues. This xylan exhibited macrophage, splenocyte, and thymocyte stimulatory activities. In vitro antioxidant studies demonstrated that the xylan has DPPH radical scavenging potential (EC50 = 148 µg/mL), ABTS radical quenching activity (EC50 = 188 µg/mL) and ferrous ions chelating activity (EC50 = 370 µg/mL). These findings elicit the need for further exploration of the xylan as a natural antioxidant and a potent immunostimulating agent.

The efficiency of using small, non-lanthanide and readily available molecules was tested as luminescent markers for gunshot residue (GSR) analysis. Three luminophores, namely pyrene (Py), fluorescein (Fl) and a Pt-CNN complex (Pt-C) were used in the present study by using their 5 wt% additives to gunpowder filled in a cartridge followed by firing with a 9 mm pistol. The easy visualization of GSR location, collection of GSR samples followed by their identification through various characterization techniques and the possibility of strategically using these markers as a cost-effective alternative compared to any lanthanide material were evaluated through this work. The comparison of physical mixture of marker and gunpowder and surface soaked gunpowder with the same marker (Py) was also evaluated. Spectroscopic (Optical, Luminescence and Raman), Microscopic, Spectrometric (Mass) and thermal analysis (Differential Scanning Calorimetry, DSC) of the marker, gunpowder and GSR residues implies that both the dyes and gunpowder retains their individual properties and those are merely a physical mixture. Overall, the present study clearly demonstrates that these small organic/metal complex based luminophores are cost effective luminescent marker as compared to lanthanide materials for GSR detection / collection and can be strategically use to track the illegal / unauthorized use of gunpowder.

The emergence of the novel coronavirus, SARS-CoV-2 has pushed forward the world to experience the first pandemic of this century. Any specific drug against this RNA virus is yet to be discovered and presently, the COVID-19 infected patients are being treated symptomatically. During the last few decades, a number of polysaccharides with potential biological activities have been invented from Indian medicinal plants. Many polysaccharides, such as sulfated xylomannan, xylan, pectins, fucoidans, glucans, glucoarabinan, and arabinoxylan from Indian medicinal plants, have been shown to exhibit antiviral and immunomodulating activities. Plant polysaccharides exhibit antiviral activities through interference with the viral life cycle and inhibition of attachment of virus to host cell. Intake of certain immune stimulating plant polysaccharides may also protect from the virus to a certain extent. In process of continuous search for most potent drug, Indian plant polysaccharides may emerge as significant biomaterial to combat COVID-19. This review explores a number of polysaccharides from Indian medicinal plants which showed antiviral and immunomodulating activities. It is aimed to provide an overview about the composition, molecular mass, branching configuration and related bioactivities of polysaccharides which is crucial for their classification as possible drug to induce immune response in viral diseases.

Mushrooms are renewable natural gift for humankind, furnished with unique taste, flavor and medicinal properties. For the last few decades study of mushroom polysaccharides has become a matter of great interest to the researchers for their immunomodulating, antimicrobial, antioxidant, anticancer, and antitumor properties. Molecular mass, branching configuration, conformation of polysaccharides and chemical modification are the major factors influencing their biological activities. The mechanism of action of mushroom polysaccharides is to stimulate T-cells, B-cells, natural killer cells, and macrophage dependent immune responses via binding to receptors like the toll-like receptor-2, dectin-1. The present review offers summarized and significant information about the structural and biological properties of mushroom polysaccharides, and their potential for development of therapeutic materials.

4-iodophenylboronic acid (IPBA) ligated luminescent gold cluster was synthesized by mixing an aqueous solution of IBPA and polyvinylpyrrolidone stabilized gold cluster (Au:PVP) in water at room temperature through chemisorption of iodine on gold nano surface. Transmission Electron microscopy (TEM) and matrix assisted laser desorption ionization (MALDI) analysis revealed that the size of these Au-clusters (1.4±0.2 nm) remain unchanged without any noticeable aggregation during synthesis. Owing to the formation of excimer between aryl moieties grafted over Au surface, the cluster exhibit strong emission peak at 335 nm. This luminescent gold cluster is used for sensing different saccharides in water at physiological pH through quenching of excimer emission peak. This strong excimer emission is significantly quenched in presence of saccharides through interaction with boronic acid moieties. The selectivity for different saccharides follows the order: fructose > galactose > maltose > glucose ~ ribose > sorbitol with hight affinity for fructose (KSV = 1.54 × 104 M-1) with Limit of Detection (LOD) of 100 µM.

This study aimed to investigate the molecular characterization, antioxidant activity in vitro, cytotoxicity study of an exopolysaccharide isolated from Citrobacter freundii. Firstly, the culture conditions were standardized by the Design of experiments (DoE) based approach, and the final yield of thecrude exopolysaccharide was optimized at 2568 ± 169 mg L-1. One large fraction of exopolysaccharide was obtained from the culture filtrate by size exclusion chromatography and molecular characteristics were studied. A new mannose rich exopolysaccharide (Fraction-I) with average molecular weight ~ 1.34 × 105 Da was isolated. The sugar analysis showed the presence of mannose and glucose in a molar ratio of nearly 7:2 respectively. The structure of the repeating unit in the exopolysaccharide was determined through chemical and 1D/2D- NMR experiments as: Finally, the antioxidant activity, and the cytotoxicity of the exopolysaccharide were investigated and the relationship with molecular properties was discussed as well.

A new water soluble galactoglucan with apparent molecular weight ~1.61 × 105 Da, was isolated from the edible mushroom Pleurotus djamor by hot water extraction followed by purification through dialysis tubing cellulose membrane and sepharose 6B column chromatography. The sugar analysis showed the presence of glucose and galactose in a molar ratio of nearly 3:1 respectively. The structure of the repeating unit in the polysaccharide was determined through chemical and NMR experiments as: In vitro antioxidant studies showed that the PDPS exhibited hydroxyl radical scavenging activity (EC50 = 1.681 ± 0.034 mg/mL), DPPH radical scavenging activity (EC50 = 3.83 ± 0.427 mg/mL), reducing power (EC50 = 4.258 ± 0.095 mg/mL), and ABTS radical quenching activity (EC50 = 0.816 ± 0.077 mg/mL). So, PDPS should be explored as a natural antioxidant.

A water soluble polysaccharide (PGPS) with molecular weight ~ 1.4 × 105 Da was isolated by alkali treatment from an edible mushroom Polyporus grammocephalus and purified by gel chromatography using sepharose-6B column. Monosaccharide analysis revealed that PGPS was made up of glucose only. PGPS contained (1 â†’ 3)-α-D-Glcp and (1 â†’ 4)-α-D-Glcp moieties in a molar ratio of nearly 1:2. Through a series of chemical and spectroscopic (1D/2D NMR) investigations, the repeating unit of the glucan was established as: →3)-α-D-Glcp(1 â†’ [4)-α-D-Glcp(1]2→ This α-glucan was observed to stimulate some prime components of immune system, namely, macrophages, splenocytes, and thymocytes.

By attaching the1-pyreneiodide ancillary ligand to the surface of polyvinylpyrrolidone-stabilized gold (Au:PVP) cluster or the cetyltrimethyl ammonium bromide-stabilized gold (Au:CTAB) nanorod, a new class of luminescent mixed ligand-stabilized gold nanostructures is synthesized. This postsynthetic surface modification method followed by us is a comparatively easier and hassle-free technique to acquire surface-active luminescent "functional nanomaterials". Careful analyses of transmission electron microscopy images revealed that the sizes of these Au-clusters or Au-nanorods remain unchanged without any noticeable aggregation in the medium. Owing to the formation of an excimer within the neighboring pyrenes mounted on the surface of core nanostructures (i.e., Au:PVP nanocluster and Au:CTAB nanorod), the resulting pyrene-grafted nanocomposites exhibit strong emission characteristics. The strong excimer emission is significantly quenched in the presence of electron-deficient chemical inputs, and this phenomenon can be used for analytical purposes. Using these luminescent Au-nanomaterials, we demonstrate a selective detection and sensing of trace-level nitroaromatic explosives (e.g., trinitrotoluene, trinitrophenol (TNP), dinitrotoluene, 4-nitrotoluene, etc.). It was observed that the Py-Au:PVP nanocluster is equally effective for explosive detection in both solution and solid phases with the limit of detection up to 10 nanomolar. A high Stern-Volmer constant of up to 3.88 × 106 M-1 was seen in the case of TNP in anhydrous methanol at 298 K. The deactivation pathway operating within the Py-Au:PVP nanocluster and the analytes is thought to be a result of a predominating static quenching process, where a nonfluorescent D-A supramolecular adduct is formed in the medium. Py-Au:PVP has also been successfully used to develop latent fingerprints from nonporous surfaces under an exposure of 365 nm UV light. The results suggest that these new composite materials could behave as potential "functional nanomaterials", which might be a promising alternative for on-the-spot detection of explosive traces as well as for easy visualization of latent fingerprints.

A green synthesis of silver nanoparticles was synthesized by AgNO3 with arabinoxylan, isolated from green stem of Andrographis paniculata (Kalmegh). The synthesized Ag NPs-arabinoxylan conjugates were characterized by UV-vis spectroscopy, FE-SEM, TEM, XRD, TGA, EDX, and Zeta potential experiments. The Ag NPs formation was established by the surface plasmon resonance band ~410.25 nm. SEM image showed mostly spherical morphology of Ag NPs. The fcc crystalline nature was identified by XRD, SAED and the size were 24.5 and 25 nm from TEM and XRD analysis respectively. The prepared Ag NPs showed dose-dependent antimicrobial activity against Streptococcus pneumonia, Candida albicans and E. coli. The nanoparciles damage 4% hemolysis to human RBCs at 12.5 µg/mL. MTT assay of Ag NPs showed that half of the cell killed at 10 µg/mL and wound healing assay observed effective inhibition cell proliferation.

The COVID-19 pandemic is considered as the most crucial global health calamity of the century and the greatest challenge that the humankind faced since the 2nd World War. In December 2019, a new infectious respiratory disease emerged in Wuhan, Hubei province, China and was named by the World Health Organization as COVID-19 (coronavirus disease 2019). A new class of corona virus, known as SARS-CoV-2 (severe acute respiratory syndrome coronavirus 2) has been found to be responsible for occurrence of this disease. As far as the history of human civilization is concerned there are instances of severe outbreaks of diseases caused by a number of viruses. According to the report of the World Health Organization (WHO as of April 18 2020), the current outbreak of COVID-19, has affected over 2164111 people and killed more than 146,198 people in more than 200 countries throughout the world. Till now there is no report of any clinically approved antiviral drugs or vaccines that are effective against COVID-19. It has rapidly spread around the world, posing enormous health, economic, environmental and social challenges to the entire human population. The coronavirus outbreak is severely disrupting the global economy. Almost all the nations are struggling to slow down the transmission of the disease by testing & treating patients, quarantining suspected persons through contact tracing, restricting large gatherings, maintaining complete or partial lock down etc. This paper describes the impact of COVID-19 on society and global environment, and the possible ways in which the disease can be controlled has also been discussed therein.

Environmentally benign mechanochemistry-assisted high-yielding synthesis of fluorescein-phenylalaninol (FPA) conjugates as a Schiff base receptor is reported herein. This newly synthesized fluorescent probe is found to be most exciting and efficient because of its simultaneous detection and removal of mercury ions (Hg2+) in aqueous medium and industrial effluents through precipitate formation. The receptor successfully worked as a chemosensor in selectively sensing the Hg2+ ion through the rapid transition from yellow to pink in the colorimetric as well as quenching of fluorescence intensity in the fluorometric assay. The removal of mercury ions was confirmed by the inductively coupled plasma analysis of the supernatant. The lower detection limit of Hg2+ ions for the receptor FPA is 1.65 and 0.34 µM as determined through absorption and fluorescence spectroscopic methods, respectively. The high removal efficiency (∼98%) of the mercury ions is promising and could be achieved via the formation of the complex in a 1:1 stoichiometric ratio of receptor to Hg2+ ions. Furthermore, this probe may be a practical alternative for use in a paper-based portable device for achieving on-site detection of mercury ions in solid, solution, and vapor phases.

A water soluble heteroglycan (THPS) of an average molecular weight ~1.98 × 105 Da was isolated from the aqueous extract of the fruit bodies of an edible mushroom Termitomyces heimii. Structural characterization of THPS was carried out using acid hydrolysis, methylation analysis, periodate oxidation, Smith degradation and 1D/2D NMR studies. Sugar analysis indicated the presence of glucose, mannose, galactose, and fucose in a molar ratio of nearly 6:2:2:1. The repeating unit of the THPS had a backbone consisting of four (1 â†’ 3)-ß-d-glucopyranosyl, one (1 â†’ 6)-ß-d-glucopyranosyl, two (1 â†’ 3)-α-D-manopyranosyl, and two (1 â†’ 6)-α-D-galactopyranosyl residues, out of which one (1 â†’ 3)-ß-d-glucopyranosyl residue was branched at O-6 position with terminal ß-d-glucopyranosyl residue and one (1 â†’ 6)-α-D-galactopyranosyl residue was branched at O-2 position with terminal α-L-fucopyranosyl residue.

RESUMO

The photoluminescence properties of aryl iodides chemisorbed on gold nanoparticles were examined. Chemisorption of a series of aryl iodides onto poly(N-vinylpyrrolidone)-protected Au nanoparticles (Au:PVP) in solution resulted in highly enhanced luminescence. Kinetic studies revealed that the photoluminescence is derived from the excimer formation of aryl iodides on Au:PVP, and the process is significantly faster than intersystem crossing by the heavy atom.

A water insoluble ß-glucan (PS), with molecular mass ∼9.16 × 104 Da was isolated from the 4% alkaline extract of an edible mushroom, Pleurotus djamor and found to consist of (1→3)-ß-d-glucopyranosyl moiety. The structure of the PS was elucidated on the basis of total hydrolysis, methylation analysis, periodate oxidation, and NMR experiments (1H, 13C, DQF-COSY, DEPT-135, and HSQC). The structure of the repeating unit of the polysaccharide was established as: →3)-ß-d-Glcp-(1→. The water insoluble ß-glucan showed cytotoxic effect against PA1 cells, where˜50% population was destroyed at 100 µg/mL concentration, and almost all cells at 250 µg/mL concentration. The wound healing assay showed significant anticarcinogenic effect against ovarian carcinoma PA1 cells after 48 h of treatment.

RESUMO

A water soluble arabinoxylan (APPS), with a molecular weight ∼1.49 × 105 Da was isolated from the green stem of Andrographis paniculata. The APPS contained D-xylose, 2-methoxy D-xylose and L- arabinose in molar ratio of 3:1:1. The structure of the repeating unit in the polysaccharide was determined through several chemical (acid hydrolysis, Methylation analysis and periodate oxidation) and spectroscopic analysis (1D/2D NMR experiments) as: In vitro antioxidant assay, APPS was found to possess ferrous ion chelating activity (EC50 = 283 µg/ml), superoxide radical scavenging activity (EC50 = 470 µg/ml), and hydroxide radical scavenging activity (EC50 = 193 µg/ml). Thus, APPS could be used as a natural antioxidant agent for food and pharmaceutical industries.
